產(chǎn)品介紹 Osteopontin is the principal phosphorylated glycoprotein of bone and is expressed in a limited number of other tissues including dentine. Osteopontin is produced by osteoblasts under stimulation by calcitriol and binds tightly to hydroxyapatite. It is also involved in the anchoring of osteoclasts to the mineral of bone matrix via the vitronectin receptor, which has specificity for osteopontin. Osteopontin is overexpressed in a variety of cancers, including lung, breast, colorectal, stomach, ovarian, melanoma and mesothelioma.

骨橋蛋白(osteopontin)又稱為早期T淋巴細(xì)胞活化因子I或分泌型磷蛋白I,是一種重要的多功能細(xì)胞外基質(zhì)蛋白,在骨組織細(xì)胞、巨噬細(xì)胞、平滑肌細(xì)胞、內(nèi)皮細(xì)胞及上皮細(xì)胞等多種細(xì)胞上表達(dá),在介導(dǎo)細(xì)胞的趨化、聚集、黏附、增殖和遷移,以及骨組織的礦化與重建、免疫調(diào)節(jié)、信號(hào)轉(zhuǎn)導(dǎo)和對感染性疾病的免疫能力等方面有著重要的作用。
OPN在細(xì)胞免疫中的功能:1.OPN對炎性細(xì)胞的作用:炎癥是炎性細(xì)胞(單核巨噬細(xì)胞、淋巴細(xì)胞、中性粒細(xì)胞等)在炎癥部位的聚集,適度的炎癥對機(jī)體有益,如抗感染及促進(jìn)組織細(xì)胞功能恢復(fù)等;但過度的炎癥能引起組織損傷等有害作用。OPN在炎癥中具有促炎及抗炎雙重作用;2.OPN對淋巴細(xì)胞的作用:早期的報(bào)道稱OPN是T淋巴細(xì)胞抑制因子,它可抑制T淋巴細(xì)胞而促進(jìn)B淋巴細(xì)胞及肥大細(xì)胞的分化、增殖
骨橋蛋白(osteopontin,OPN)是一種含精氨酸—甘氨酸—天冬氨酸(Arg-Gly-Asp,RGD)的分泌型糖基化磷蛋白,已歸類于細(xì)胞外基質(zhì)(extracellular matrix,ECM)蛋白。 OPN既是細(xì)胞因子,又是細(xì)胞黏附分子。
OPN最初是從骨組織分離的,而后在上皮細(xì)胞、胃腸道的分泌物、腎、甲狀腺、睪丸以及一些腫瘤細(xì)胞里都陸續(xù)的發(fā)現(xiàn)了OPN的表達(dá)。在生殖方面,OPN在分泌期子宮內(nèi)膜上皮細(xì)胞、侵襲的滋養(yǎng)細(xì)胞、蛻膜細(xì)胞中表達(dá)。

Paraformaldehyde-fixed, paraffin embedded (Rat lung); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with (Osteopontin) Polyclonal Antibody, Unconjugated (bs-0026R) at 1:400 overnight at 4°C, followed by a conjugated secondary (sp-0023) for 20 minutes and DAB staining.

Secondary Antibody : Goat anti-rabbit IgG-AF647 Dilution: 1μg /test. Protocol The cells were fixed with 4% PFA (10min at room temperature)and then permeabilized with 0.1% PBST for 20 min at room temperature. The cells were then incubated in 5%BSA to block non-specific protein-protein interactions for 30 min at room temperature .Cells stained with Primary Antibody for 30 min at room temperature. The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed.
