background: The product of this gene functions in the nucleotide excision repair pathway, and is required for the repair of DNA lesions such as those induced by UV light or formed by electrophilic compounds including cisplatin. The encoded protein forms a heterodimer with the XPF endonuclease (also known as ERCC4), and the heterodimeric endonuclease catalyzes the 5' incision in the process of excising the DNA lesion. The heterodimeric endonuclease is also involved in recombinational DNA repair and in the repair of inter-strand crosslinks. Mutations in this gene result in cerebrooculofacioskeletal syndrome, and polymorphisms that alter expression of this gene may play a role in carcinogenesis. Multiple transcript variants encoding different isoforms have been found for this gene. The last exon of this gene overlaps with the CD3e molecule, epsilon associated protein gene on the opposite strand. ERCC1蛋白是動物內(nèi)重要的DNA修復基因家族成員之一。在人體內(nèi)它的低表達往往伴隨著基因不穩(wěn)定性增加,從而產(chǎn)生腫瘤惡性變的發(fā)生。ERCC1是損傷DNA核苷酸切除修復中高度保守的切除性核酶。在哺乳動物的核苷酸切除修復中，XPG負責切割DNA損傷區(qū)域的3’端，而ERCC1-XPF復合物在5’端打開切口。目前初步的研究表明，癌瘤中的ERCC1蛋白表達程度越低，順氯氨鉑化療的效果就越好。ERCC1蛋白陽性的患者，非化療治療的預后要好于接受化療。 |
